Establishing a suitable detection method for residual DNA in host cells is crucial for monitoring the safety and quality controllability of biological products. Among them, the qPCR method has become the preferred detection method for process research and product quality control of biological products because of its high sensitivity, specificity, accuracy and durability.
Residual host cell DNA in biological preparations (recombinant protein drugs, antibody drugs, vaccines, cell therapy products, etc.) may pose infectious or tumorigenic risks, and it is necessary to control the residual DNA of host cells to an acceptable level. Host cell residual DNA testing can ensure that biological preparations meet the requirements of regulatory agencies such as China, WHO, Europe, and the United States. It is recommended to conduct host cell DNA residue research on process samples, and put the host cell DNA residue detection into the stock solution or preparation release inspection. The 2020 edition of "Chinese Pharmacopoeia" Part 3 - 3407 Exogenous DNA Residue Determination Method includes the qPCR method for the detection of host residual DNA in biological products. This method has the characteristics of rapidity, high throughput, and high sensitivity.
